There is a chariot stop located in the center of every city center. Each chariot stop is linked to several other stops in adjacent regions. By using the chariots, you may instantly travel to any of the linked stops.
Travel from one stop to another normally requires offline travel time, which is collected while you are logged off. Periodically, however, the route from one stop to another will become free for two minutes. You may view the cost in travel time and the time until the next free ride at the chariot stop.
For example, a stop might say:
Travel to Nile Delta requires 2 hours 40 minutes 55 seconds of travel time.
Travel will be free in 5 minutes
In this case, you may either spend the 2 hours, 40 minutes, and 55 seconds of offline travel time to instantly move to the Nile Delta, or wait about 5 minutes for a free ride.
The offline travel time needed for a journey is proportional to the distance as the crow flies. So the most direct (physically shortest) route is also the least expensive route in terms of offline travel time if you're in a hurry.
1. Many people refer to the actual "chariot" that is arriving as a "bus". 2. There is actually one more minute to wait than it says there is. When it says "1 minute remaining" it will count down one minute and THEN count down 60 seconds. So, if there are 12 minutes remaining, there are actually 13.
There is now a predicted Chariot Schedule and route planner created by Tialaramex. This lets you discover when it will be possible to ride for free without actually visiting the chariot stop itself.
(The same planner with a different front system is here).

Don't know if this helps, but this is route I used to visit all the Universities of Worship when getting my seeds.
Sinai -> Nile Delta -> Pharaoh's Bay -> Valley of Kings -> Fools Paradise -> Desert of Shades -> Red Sea Oasis -> Karnak -> Lower Nubia -> Upper Nubia -> Cradle of the Sun -> Sinai
By starting wherever you need to, this route takes you to each of the chariot stops available.
Hopefully later I'll be able to work out a more efficient route in terms of offline travel time - but no promises!
When a Megalopolis is built, it upgrades the nearby? chariot stop with a message board. Chariot stops upgraded so far are listed on the Test of the Megalopolis page.
